This book is a Win for those in the HR and training profession on how to use the scoreboard methods in training. The chapter on the Value of Training Scorecards by Ms Schmidt is a must read. The case studies by those in the field are great examples of how to apply the concepts. This book should be in everyones reference book shelf. Jack Phillips and Lynn Schmidt are a great team in presenting concepts of ROI and value of training scorecards. Thanks to the editors and the contributors for providing this valuable resource. I have many years of experience in training and management development from postions in telecom and computers.

Beautiful eclipse photography.......2001-02-15
I have an extensive collection of eclipse books, and this one really stands out in the crowd. For one thing, it's easily the largest-format eclipse book I've seen; I'd call it a coffee-table book. But what is really striking are the beautiful photographs. The reproduction is outstanding, and the large size makes the photos particularly vivid.
I haven't read the text yet, and so I can't comment on it, but the overall impression is that this book is a must-have for anybody interested in the beauty of total solar eclipses.

To examine the potential of a suite of biomarkers as early warning indicators of environmental pollution, sperm count, neutral red retention time (NRRT) and DNA damage were measured in earthworm Eisenia fetida exposed to increasing concentrations of acetochlor in OECD soil. The neutral red retention time of earthworms coelomocytes was sensitive to acetochlor pollution, and decreased significantly when the concentration was more than 10mgkg^-^1 after 30 and 60 days of exposure (P
<0.05). The reduced neutral red retention time correlated with the soil acetochlor residual. Sperm count decreased significantly at the concentrations of 40 and 80mgkg^-^1 after 15 days of exposure (P
<0.05). The DNA damage of earthworms coelomocytes increased significantly after 30 days of exposure at the highest concentration (80mgkg^-^1; P
<0.05). Earthworms were under physiological stress at field dose of acetochlor (10mgkg^-^1). Higher concentrations of acetochlor caused sperm count decrease and DNA damage of earthworms. Such a suite of biomarkers could serve as indicators of the health of the soil environment and to evaluate the toxicity of acetochlor on earthworms or as a means of monitoring soil acetochlor pollution.

The heavy metal cadmium (Cd^2^+) applied on tobacco roots in the form of cadmium chloride, induced significantly higher levels of DNA damage as measured by the cellular Comet assay than did treatment of isolated root nuclei, analyzed by use of the acellular Comet assay. DNA damage induced by Cd^2^+ in roots of a transgenic catalase-deficient tobacco line (CAT1AS) was higher than in wild-type tobacco (SR1) roots. In contrast to treatment with the positive control ethyl methanesulphonate, Cd^2^+ induced no significant DNA damage in leaf nuclei, and neither somatic mutations, nor homologous recombination as measured by the GUS genereactivation assay, were observed in leaves. Analysis of the accumulation of cadmium by inductively coupled plasma optical emission spectrometry demonstrates that roots accumulate almost 50-fold more cadmium than above-ground parts of the tobacco seedlings. This may explain the absence of Cd^2^+ genotoxicity in leaves.

Investigations of inorganic oligomers are important in both chemistry and physiology. In this contribution, we propose a laser induced light scattering imaging (LSI) and a total internal reflected light scattering imaging (TIR-LSI) technique, and apply them to characterize the interactions of inorganic oligomers with biopolymer in aqueous phase and at liquid/liquid interface, respectively. In aqueous medium, synthetic chromium(III) hydrolytic oligomers (CrHO) react with DNA, and the resultant binary could be extracted into the H"2O/CCl"4 interface in the presence of triocyctyl phosphine oxide (TOPO), forming a DNA-CrHO-TOPO ternary amphipathic complex at the interface with the associate constant of 1.32x10^3mol^-^1dm^4 for a given 1.0x10^-^4moll^-^1 TOPO. Under the excitation of a 441-nm He-Cd laser light beam, the resultant light scattering and total internal reflected light scattering (TIR-LS) signals of the formed binary in aqueous phase and ternary at liquid/liquid interface could be easily captured using a common microscope coupled with a CCD camera. By digitally analyzing the CCD captures, we demonstrate that aggregations of the CrHO-DNA binary in aqueous phase and DNA-CrHO-TOPO ternary at liquid/liquid interface have occurred, respectively.

It has been reported that the phosphorylated form of histone variant H2AX (@cH2AX) plays an important role in the recruitment of DNA repair and checkpoint proteins to sites of DNA damage, particularly at double strand breaks (DSBs). Using @cH2AX foci formation as an indicator for DNA damage, several chemicals/stress factors were chosen to assess their ability to induce @cH2AX foci in a 24h time frame in a human amnion FL cell line. Two direct-acting genotoxins, methyl methanesulfonate (MMS) and N-ethyl-N-nitrosourea (ENU), can induce @cH2AX foci formation in a time- and dose-dependent manner. Similarly, an indirect-acting genotoxin, benzo[a]pyrene (BP), also induced the formation of @cH2AX foci in a time- and dose-dependent manner. Another indirect genotoxin, 2-acetyl-aminofluorene (AAF), did not induce @cH2AX foci formation in FL cells; however, AAF can induce @cH2AX foci formation in Chinese hamster CHL cells. Neutral comet assays also revealed the induction of DNA strand breaks by these agents. In contrast, epigenetic carcinogens azathioprine and cyclosporine A, as well as non-carcinogen dimethyl sulfoxide, did not induce @cH2AX foci formation in FL cells. In addition, heat shock and hypertonic saline did not induce @cH2AX foci. Cell survival analyses indicated that the induction of @cH2AX is not correlated with the cytotoxic effects of these agents/factors. Taken together, these results suggest that @cH2AX foci formation could be used for evaluating DNA damage; however, the different cell types used may play an important role in determining @cH2AX foci formation induced by a specific agent.

Toxicogenomics represents the merging of toxicology with genomics and bioinformatics to investigate biological functions of genome in response to environmental contaminants. Aquatic species have traditionally been used as models in toxicology to characterize the actions of environmental stresses. However, since microarray technology is thus far limited to only a few aquatic species and derivation of biological meaning from microarray data is highly dependent on statistical arguments, the full potential of microarray in aquatic species research has yet to be realized. Herein we review some of the issues related to construction, probe design, statistical and bioinformatical data analyses, and current applications of DNA microarrays. As a model a recently developed medaka (Oryzias latipes) oligonucleotide microarray was described to highlight some of the issues related to array technology and its application in aquatic species exposed to hypoxia. Although there are known non-biological variations present in microarray data, it remains unquestionable that array technology will have a great impact on aquatic toxicology. Microarray applications in aquatic toxicogenomics will range from the discovery of diagnostic biomarkers, to establishment of stress-specific signatures and molecular pathways hallmarking the adaptation to new environmental conditions.

We compared the DNA damaging potency of acrylamide (AA) and its metabolite glycidamide (GA) in the comet assay in cell systems differing with respect to species origin and cytochrome P450-depended monooxygenase (CYP2E1) expression (V79, Caco-2, primary rat hepatocytes). Only after 24h incubation in the highest concentration of AA (6mM) a slight but significant increase in DNA damage was observed in V79 and Caco-2 cells. In primary rat hepatocytes, however, expressing substantial amounts of CYP2E1, no induction of DNA strand breaks was found. At the end of the incubation time period (24h), still 67+/-19% of the CYP2E1 protein was detected by Western blotting. Direct treatment with GA resulted in a significant increase in DNA damage in V79 cells and primary rat hepatocytes at concentrations >=100@mM (24h). Caco-2 cells were found to be less sensitive, exhibiting an increase in DNA strand breaks at concentrations >=300@mM GA. These data confirm the higher genotoxic potential of GA compared to AA but also indicate that high expression of CYP2E1 per se is not necessarily associated with increased genotoxicity of AA. We, therefore, investigated whether the intracellular glutathione (GSH) level might be a critical determinant for the genotoxicity of AA in cells with different CYP2E1 status. Depletion of intracellular GSH by dl-buthionine-[S,R]-sulfoxime (BSO) in rat hepatocytes and V79 cells resulted in a significant induction of DNA strand breaks after incubation with 1mM AA. However, at higher concentrations (>=1.25mM) a strong increase in cytotoxicity, resulting in a severe loss of viability, was observed. In summary, the DNA strand breaking effect of AA appeared not to be directly correlated with the CYP2E1 status of the cells. Depletion of GSH is associated with an increase in AA genotoxicity but seems also to lead to a substantial enhancement of cytotoxicity.

RecA protein plays a central role in homologous recombination and DNA repair. RecX, a gene directly downstream in Escherichia coli and some other bacterial species, down regulates it. However, the precise mechanism of regulation of RecA by RecX is not known. In order to study the function of RecX in the highly radioresistant bacterium Deinococcus radiodurans, null and overexpression strains were constructed. Our data demonstrates that RecX represses radiation resistance, DNA recombination and consequent genome instability in the stationary phase bacteria. Further biochemical analyses reveal that RecX not only down regulates recA transcription, but also directly inhibits RecA activities in vitro. These data suggests a dual negative regulatory control of RecX on RecA functions in D. radiodurans.

The preparation and detection of highly luminescent aqueous europium nanoparticle labels for the time-resolved fluorometric assays has been demonstrated. The luminescent complexes included in the nanoparticle were composed of a dye sensitizer and a polyaminocarboxylate-based chelator with excellent water-solubility and a high binding constant for lanthanides. The luminescence of europium is greatly enhanced by the sensitization of the dye. The structure design of two functional entities in the single molecule made the lanthanide complex to have both strong luminescence and good aqueous solubility. SiO"2 nanoparticles containing europium complexes were prepared by the reverse microemulsion method. One particle of ~55nm in diameter has a luminescent intensity corresponding ~1300 free europium complexes and 0.61ms fluorescence lifetime. The DNA time-resolved fluorometric assay with this nanoparticle label was carried out by magnetic microbeads as solid phase carrier. The detection sensitivity with the nanoparticles is improved more than 50-fold compared with the conventional dye FITC molecules. This luminescent nanoparticle label based on dye sensitization can be applied in ultrasensitive time-resolved fluorometric assays and imaging.

DNA lesions can stall or block high-fidelity polymerases, thus inhibiting replication. To bypass such lesions, low-fidelity translesion synthesis (TLS) polymerases can be used to insert a nucleotide across from the lesion or extend from a lesion:base mispair. When DNA repair is compromised in Saccharomyces cerevisiae, spontaneous DNA lesions can lead to a novel mutational event in which a frameshift is accompanied by one or more base pair substitutions. These ''complex frameshifts'' are dependent upon the TLS polymerase Pol@z, and provide a mutational signature for mutagenic Pol@z-dependent activity. In the current study, we have found that a specific subset of the Pol@z-dependent mutational events requires oxidative metabolism. These results suggest that translesion bypass of spontaneously oxidized DNA bases can be a significant source of mutagenesis in repair compromised cells.
