Well-Grounded Advice.......2002-07-11
Many managers never seek to refine their management style. Subconsciously, they feel that their "many years of experience" or innate intelligence has rightfully propelled them to their current position. For other managers, who constantly strive to improve their performance in the workplace, this book is for you.
At first glance, the topics covered may seem intuitive. Readers may initially shun chapters that underscore the importance of ethics, employee independence, and worker satisfaction, dismissing them as old hat and self-explanatory. Upon further inspection however, the unsuspecting reader will discover that the book's true power lies in its explicit detail of the benefits associated with these seemingly well-trodden concepts. The book does an exceptional job of weaving the topics back into a bigger picture of organizational success.

Strong scientific case for artificiality.......2000-07-21
Carlotto brings state of the art image analysis techniques to bear on the enigmatic objects of Mars' Cydonia region. He shows that there is a high degree of probability for artificiality, something that NASA and Michael Malin vigorously deny - to this day. This book is a must-read for any serious student of the artificiality hypothesis.

Mark Carlotto's book is a must-buy. Every page is a discovery in itself; the author makes his case, and makes it extremely well: Mars is probably home to non-natural features well within our technological ability to investigate (preferably through manned exploration).
"The Martian Enigmas" is a satisfying piece of popular science that effectively sounds the death toll for "skeptics" who insist it's all our imaginations.
Superb title for Mars Images and Advanced Image Processing.......1999-01-14
This book explores the fundamentals of image processing as applied to satellite photography. The author also details the processing of the images of constructed features on the surface of the planet Mars. Mr. Carlotto was able to demonstrate in very easy terms the methods used to illuminate these breathtaking images. I eagerly await any book from this author on the latest set of images taken in 1998.

The study described in the present article used fluorescence resonance energy transfer (FRET) to monitor the folding of a 31-mer cytosine-rich DNA segment, from the promoter region of the human c-myc oncogene. Spectroscopic FRET data recorded during experiments carried out in different experimental conditions were individually and simultaneously analyzed by multivariate curve resolution. The simultaneous analysis of several data matrices allowed the resolution of the system, removing most of the ambiguities related to factor analysis. From the results obtained, we report the evidence of the formation of two ordered conformations in acidic and neutral pH values, in addition to the disordered structure found at high temperatures. These ordered conformations could be related to cytosine-tetraplex structures showing different degrees of protonation in cytosine bases.

Nanoparticles containing thousands of fluorescent europium(III) chelates have a very high specific activity compared to traditional lanthanide chelate labels. It can be assumed that if these particles are used in a homogeneous assay as donors, multiple chelates can excite a single acceptor in turns and the energy transfer to the acceptor is increased. The principle was employed in an immunoassay using luminescent resonance energy transfer from a long lifetime europium(III) chelate-dyed nanoparticle to a short lifetime, near-infrared fluorescent molecule. Due to energy transfer fluorescence lifetime of the sensitised emission was prolonged and fluorescence could be measured using a time-resolved detection. A competitive homogeneous immunoassay for estradiol was created using 92nm europium(III) chelate-dyed nanoparticle coated with 17@b-estradiol specific recombinant antibody Fab fragments as a donor and estradiol conjugated with near-infrared dye AlexaFluor 680 as an acceptor. The density of Fab fragments on the surface of the particle influenced the sensitivity of the immunoassay. The optimal Fab density was reached when the entire surface of the particle participated in the energy transfer, but the areas where the energy was transferred to a single acceptor, did not overlap. We were able to detect estradiol concentrations down to 70pmoll^-^1 (3xSD of a standard containing 0nmoll^-^1 of E2) using a 96-well platform. In this study we demonstrated that nanoparticles containing lanthanide chelates could be used as efficient donors in homogeneous assays.

Dual-labeled oligonucleotide derivative, FAT-0, carrying 6-carboxyfluorescein (FAM) and 6-carboxy-tetramethylrhodamine (TAMRA) labels at 5'- and 3'-termini of thrombin-binding aptamer (TBA) sequence 5'-GGTTGGTGTGGTTGG-3' and its derivatives, FAT-n (n=3, 5, and 7) were designed and synthesized. FAT-n derivatives contained a T"mA spacer (m=2, 4, and 6, respectively) at 5'-end of TBA sequence. The probes were developed to estimate the spacer effect on FRET efficiency and to identify the best probe for sensing of K^+. Circular dichroism (CD), UV-vis absorption, and fluorescence studies revealed that all FAT-n probes could form the intramolecular tetraplex structures after binding K^+. Association constants of particular K^+/FAT-n complexes were determined using different experimental approaches. Suitability of particular probes for sensitive monitoring of K^+ in intra- and extracellular conditions was examined and discussed. Calibration graphs of fluorescence ratio were linear in the K^+ concentration range of 2-10mM for extracellular conditions showing sensitivity of 1.2%mM^-^1 K^+ and for intracellular conditions in the range of 100-200mM with sensitivity of 0.49%mM^-^1 K^+.

Fluorescence resonance energy transfer (FRET) between the extrinsic dye labels Cyanine 3 (Cy3), Cyanine 5 (Cy5), Carboxytetramethyl Rhodamine (TAMRA), Iowa Black Fluorescence Quencher (IabFQ), and Iowa Black RQ (IabRQ) has been studied. The Forster distances for these FRET-pairs in single- and double-stranded DNA conjugates have been determined. In particular, it should be noted that the quantum yield of the donors Cy3 and TAMRA varies between single- and double-stranded DNA. While this alters the Forster distance for a donor-acceptor pair, this also allows for detection of thermal denaturation events with a single non-intercalating fluorophore. The utility of FRET in the development of nucleic acid biosensor technology is illustrated by using TAMRA and IabRQ as a FRET pair in selectivity experiments. The differential quenching of TAMRA fluorescence by IabRQ in solution has been used to discriminate between 0 and 3 base pair mismatches at 60^oC for a 19 base sequence. At room temperature, the quenching of TAMRA fluorescence was not an effective indicator of the degree of base pair mismatch. There appears to be a threshold of duplex stability at room temperature which occurs beyond two base pair mismatches and reverses the observed trend in TAMRA fluorescence prior to that degree of mismatch. When this experimental system is transferred to a glass surface through covalent coupling and organosilane chemistry, the observed trend in TAMRA fluorescence at room temperature is similar to that obtained in bulk solution, but without a threshold of duplex stability. In addition to quenching of fluorescence by FRET, it is believed that several other quenching mechanisms are occurring at the surface.

A disposable sensor has been developed for the measurement of copper(II) concentration in aqueous solution based on a change in the fluorescence of porphyrazine 2,7,12,17-tetra-tert-butyl-5,10,15,20-tetraaza-21H,23H-porphine (TP). The sensor was constructed by spin-coating a polyester support with a PVC solution containing TP, a plasticizer, the chelating agent Zincon and the ion-pairing benzetonium chloride. The measurement principle is based on the radiationless resonance energy transfer (RET) from TP immobilized in membrane, and acting as fluorescence donor, to Zincon acting as an acceptor induced by copper(II). The absorption spectrum of the Zincon-Cu(II) complex presents adequate overlapping with the emission spectrum of TP, producing a useful analytical signal by the RET process. The disposable sensor responds to copper(II) irreversibly over a dynamic range from 0.039 to 14@mmolL^-^1 (2.5-890@mgL^-^1) with a sensor-to-sensor reproducibility (relative standard deviation RSD) of 1.9%, as log a"C"u"^"2"^"+, at the medium level of the range and a response time of 10min. The performance of the optical disposable sensor was tested for the analysis of copper in different types of natural waters (river, well, spring and swimming pool), validating results against a reference procedure.

Non-overlapping fluorescence resonance energy-transfer (nFRET) is a highly sensitive, time-resolved, assay technology. It utilizes lanthanide chelates as donors together with non-overlapping acceptor fluorophores, which have their absorption maximum energetically at a higher level than the emittive transitions of the donor. In this report the nFRET was studied using a low quantum yield Sm(III)-chelate as the donor in a homogeneous dF508-DNA hybridization assay. Despite the low quantum yield, Sm(III) could function as an efficient donor in nFRET, which resulted in strong energy-transfer induced acceptor emission in the assay. Further, the induced acceptor emission of the assay showed non-Forster-type decay characteristics and allowed the sensitive anti-Stokes' shift FRET measurement, in which the induced acceptor emission was measured at a wavelength region shorter than the main emittive transitions of the donor. The detection limit for the dF508 DNA-target was 12.0pM (S/B=2). The assay sensitivity was surprisingly similar with the corresponding Eu-based nFRET-assay, despite the approximately 90-fold lower quantum yield of the Sm-donor. The results confirm the exceptional properties of the nFRET induced acceptor signal and support the theory of energy-transfer taking place from the excited energy levels above the emittive energy level of the lanthanide donor. We conclude that nFRET-technique is well suited for highly sensitive DNA assays and that the assay sensitivity is not limited by the emittive quantum yield of the donor.

A homogeneous non-competitive assay principle for measurement of small analytes based on quenching of fluorescence is described. Fluorescence resonance energy transfer (FRET) occurs between the donor, intrinsically fluorescent europium(III)-chelate conjugated to streptavidin, and the acceptor, quencher dye conjugated to biotin derivative when the biotin-quencher is bound to Eu-streptavidin. Fluorescence can be measured only from those streptavidins that are bound to biotin of the sample, while the fluorescence of the streptavidins that are not occupied by biotin are quenched by quencher-biotin conjugates. The quenching efficiencies of the non-fluorescent quencher dyes were over 95% and one dye molecule was able to quench the fluorescence of more than one europium(III)-chelate. This, however, together with the quadrovalent nature of streptavidin limited the measurable range of the assay to 0.2-2nmolL^-^1. In this study we demonstrated that FRET could be used to design a non-competitive homogeneous assay for a small analyte resulting in equal performance with competitive heterogeneous assay.

Walter A. Atkinson is brilliant. Not only does he compare the simple life of the 30's, 40's and 50's with wit and the comedic wisdom of a Will Rogers, he makes the comparison with today's society and drives a conservative knive into the heart of the misguided liberal agenda. I loved this book! Atkinson had me rolling on the floor with laughter only to make me stop, reflect and realize that, yes by golly, he's right. How in the world DID we get from there to here? How did it happen in such a short period of time? Even if you can't remember what it was like during a time of elderly respect, manners, REAL movie stars, true heroes, and a grand life style even without credit cards and cell phones, you'll be transported back to those days with many a smile and an occasional grumble. There's another reason I cherish these recollections; BECAUSE I'M IN THEM!

pie-in-the-face. Every spy movie . . . used . . . the same actors to play the same characters.

of yet another sub-culture from ýhis good old days of yore.ý Forgive Us Our Senior
Moments is Atkinsonýs first book and he has fun with it. Itýs one manýs interpretation of
America--how it used to be and how it is today. The writing is droll and delightfully
sardonic, with a touch of nostalgic, old-fashioned patriotism thrown in for good measure.
One can almost feel Atkinsonýs perverse glee as he takes his forty year supply of ýprivate
gripes and wisdom pearlsý and just ýlets it rip.ý Truly a volume on senior reflection and
opinionated thought, these essays connect practically every social problem in America
today to a self-proclaimed ýcultural revolutioný of the mid-1960s.
Chapters cover ancestors, retirement, sex, music, sports, religion and lots of
politics. On retirement, and a riveting sense of impending doom, Atkinson states, ý. . . if I
had my druthers, Iýd be shouting the line Anita OýDay was singing with Gene Krupaýs
band in 1941, ý. . . just let me off uptown.ýý He talks of family and friends, and adventures

final days balancing sacred moments of joy and sorrow from the authorýs aptly described
blue ribbon years, against his ýinane, do as you please, liberal tripeý of the last several
decades. Most will relate to Atkinsonýs throwback passion for family, country and God,
and his repetitive query, ýHow did America ever get from there to here?ý
Naturally, as any thesis with a political slant will do, folks of another viewpoint
will be totally bent out of shape by much of the authorýs intended wisdom. Liberals will be
particularly upset as Atkinson hammers away with gleeful redundancy on foibles at the
heart of their core beliefs--the first and foremost being: Scare the hell out of old folks, and
keep doing it year after year, after year, after year, ad nauseam. He takes certain notable,
liberal politicians to task, citing, where apropos, their public, decadent personal lifestyles,
as well as their innate inability to fool the people-at-large if ever they should be of a mood
to posture as statesmen in public. As Atkinson states in his preface, ýWhere convictions
differ feel at liberty to consider my view a senior moment.ý

Atkinson has achieved much with this little book. It's a wonderful collection of essays which harken back to a time when things were different in America. Not for liberals, this book will thrill conservatives of whatever age.

This book addresses the dangerous, the legal technicalities, and the "seamy" side of containing prisoners. Yet Mr. Grunska describes this gauntlet with compassion, wit and humor as he tells what the guards and supervisors had to do to calm and solve the comical situations. It's a rapid-read collection of individual incidents! I loved the chapter "High Tech and Us". It reminded me of earlier years when school admimistrators gave us computers, students, and orders for "immediate use effectively."

This book was very entertaining, funny and easy to read.
The chapter about the dancing girls was hilarious and I really identified with the story about the computer mixups.

Each chapter is a funny story in itself. Who ever thought prison life was dull? This author finds the humor in every situation, bizzare as it may be. I especially liked The Dancing Girls Frolic and The Pink Panty Scenario. Who would ever suspect such shinanigans took place in such a setting?
It seems like the inmates are more clever than the guards and spend most of their time outwitting their keepers!
I highly recommend this fast paced, light reading for sheer enjoyment of a little known subject. Treat yourself to some fun.
